Description (résumé)

Ascaris suum affects feed conversion and weight gain of pigs, but effects on intestinal microbiota remain largely unexplored. Therefore, the intestinal microbiome and metabolome was assessed in pigs that were infected once with 10,000 A. suum eggs as well as pigs that received a trickle-infection (1,000 eggs/day, 10 days), compared to a control group. Six pigs per group were sacrificed at 21, 35 and 49 days post infection (dpi). Faecal samples taken weekly until slaughter as well as ingesta samples from different intestinal compartments were subjected to NGS of the bacterial 16S rRNA gene. Metabolomic profiles were assessed in ingesta and serum samples by nuclear magnetic resonance. While the single-infection caused an overall decrease in microbial diversity and a remarkable, but transient, alteration of the caecal microbiota, trickle-infection rather led to an increased microbial diversity. An increase of short chain fatty acid (SCFA) producing genera in the caecum at 21 dpi, which shifted to a decrease on day 35 pi, was a common finding in both groups, possibly related to the final moult of the parasite. Significant metabolomic changes were mainly found in the trickle-infected group, with an increase of valerate, proprionate and butyrate in the colon at 21 dpi, but a decrease at 35 dpi in the caecum and colon. In serum samples, glucose was reduced in trickle-infected pigs at 21 and 35 dpi, but increased at 49 dpi.
